Swinholide A is an actin-binding marine polyketide that dimerizes actin with a Kd of approximately 50 nM. It is a microfilament disrupting marine toxin that stabilizes actin dimers and severs actin filaments. It disrupts the actin cytoskeleton of cells and exhibits antifungal activity. Originally isolated from the Okinawan marine sponge *Theonella swinhoei*, Swinholide A is highly cytotoxic to various cancer cell lines. It sequesters actin dimers *in vitro* and rapidly severs F-actin *in vitro* with high cooperativity.
